目的建立高效液相色谱(HPLC)法同时测定中药制剂烧伤膏中5个蒽醌类化合物(芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚)的含量。方法以5个蒽醌类成分为指标,选择Diamonsil C18(4.6 mm×250 mm,5 μm)色谱柱,使用甲醇-乙腈0.1%磷酸(23∶42∶35)为流动相进行洗脱。结果5个蒽醌类成分分别在0.005 5~0.176 1 μg(芦荟大黄素r=0.999 9)、0.010 1~0.323 7 μg(大黄酸r=0.999 9)、0.016 6~0.522 3 μg(大黄素r=0.999 9)、0.0216~0.690 6 μg(大黄酚r=0.999 9)和0.030 3~0.968 8 μg(大黄素甲醚r=0.999 9)内,与峰面积呈良好的线性关系;平均回收率分别为97.6%、96.6%、98.8%、99.2%、97.5%;RSD(n=6)分别为2.0%、1.8%、2.9%、1.3%、1.2%。结论该方法简便、可靠、重复性好,适用于烧伤膏中大黄的质量控制。
Abstract
OBJECTIVE To establish a method for determining the contents of aloe-emodin, rhein, emodin, chrysophanol and physcion in the burn ointment. METHODS Diamonsil C18 column (4.6 mm×250 mm,5 μm) was adopted, with methanol-acetonitrile-0.1% phosphoric acid (23∶42∶35) as the mobile phase. The detection wavelength was set at 254 nm, the flow rate was 1 mL·min-1, and the column temperature was 30 ℃.RESULTS The liner rangers of aloe-emodin, rhein, emodin, chrysophanol and physcion were within 0.005 5-0.176 1 μg (r=0.999 9), 0.010 1-0.323 7 μg (r=0.999 9), 0.016 6-0.522 3 μg (r=0.999 9), 0.021 6-0.690 6 μg (r=0.999 9) and 0.030 3-0.968 8 μg (r=0.999 9), respectively. The average recoveries were 97.6% (RSD=2.0%),96.6% (RSD=1.8%), 98.8% (RSD=2.9%), 99.2% (RSD=1.3%), and 97.5% (RSD=1.2%), respectively (n=6). CONCLUSION This method is specific, accurate, reproducible, and can be used for the determination of aloe-emodin, rhein, emodin, chrysophanol and physcion in the burn ointment.
